Lung cancer metastasis is one of the reasons for the failure of lung cancer therapy and leads to the death of patients with malignant neoplasms. Collective tumor cell migration plays an important role in cancer metastasis. We study novel 5-cyanopyrimidine based compounds – SV296 and SV350 with better cytotoxic action on A549 lung cell line and A549 resistant to gefitinib (A549GR) by on the ability to inhibit the migration of tumor cells. To investigate the inhibition of A549/A549GR cell migration activity we performed the wound healing assay after 48 hours of cultivation with SV296, SV350 and gefitinib (Fig.1). Both synthesized compounds induce a severe inhibition of A549 cell migration compare to positive control group (65.9% and 43.5%, respectively). Gefitinib inhibited the migration activity of A549 cells with respect to the intact positive control, but did not cause significant inhibition in gefitinib-resistant cells (22.6%). The inhibitory activity of SV296 and SV350 did not significantly change in resistant cells (75.2% and 42.5%, respectively). Based on these results, it is evident that SV296 and SV350 is inhibiting cancer cell migration on gefitinib-resistant cancer cells.
