Exosomes, secreted extracellular vesicles with innate ability to transport cargo throughout the body, make them ideal candidates for drug delivery. Unfortunately, this natural ability to carry cargo is also the main obstacle to using exosomes as a drug carrier. The inherent cellular cargo of an exosome can negatively impact its ability to carry and deliver an external payload. To increase targeting and predictably control drug release, a neuropilin receptor agonist peptide and a hypoxia-responsive lipid have been incorporated into the lipid bilayer of bovine milk exosomes. Hence, peptide targeted, hypoxia-responsive bovine milk exosomes (HRX) encapsulating the anticancer drug doxorubicin or the cancer stemness inhibitor napabucasin will decrease cell survival of a heterogeneous triple-negative breast cancer cell population. Doxorubicin and napabucasin have been encapsulated using two different methods with entrapment efficiencies above 65% for both. Transmission electron microscopy, atomic force microscopy, dynamic light scattering, western blot, and qPCR have demonstrated the stability of modified exosomes in normoxic conditions and breaking under hypoxic conditions. Future studies will include testing of hypoxia-responsive exosomes’ toxicity and cellular internalization in monolayer and three-dimensional cultures before <i>in vivo</i> studies.<br/>
