Leishmania braziliensisis a protozoan parasite endemic in Brazil and other South American countries, where it causes different forms of leishmaniasis, including cutaneous and mucocutaneous leishmaniasis. Several Leishmania species (e.g., L. major, L. mexicana, and L. braziliensis) express unique Type-2 glycosylinositolphospholipids (GIPLs), which are embedded and broadly distributed on the plasma membrane of all parasite stages. These GIPLs contain highly immunogenic glycans such as α-galactopyranosides (Galpα) and/or β-galactofuranosides (Galfβ), which are absent on human cells. The detection of Galpα- or Galfβ-containing glycotopes can therefore be potentially useful in serological assays for diagnosis and/or early assessment of chemotherapeutic outcomes of leishmaniasis. Here, we demonstrate a reversed immunoglycomics approach, in which glyco-biomarkers for L. braziliensis infection are discovered by interrogating a synthetic glycoarray containing a terminal, nonreducing Galpα- or Galfβresidue in di-, tri-, or tetrasaccharide of Type-2 GIPL-1, -2, and -3 by chemiluminescent enzyme-linked immunosorbent assay (CL-ELISA), using sera from patients from two endemic regions, Bolivia and Argentina.The modular chemical synthesis of the terminal glycan partial structures of these GIPLs, their conjugation to bovine serum albumin resulting in neoglycoproteins (NGPs), and their antigenic properties studied by CL-ELISA will be presented. This work provides important insights into the molecular structures of glycan biomarkers for L. braziliensis infection in humans.
