Pectin has three primary structural domains of homogalacturanon (HG), rhamnogalacturonan I (RGI), and rhamnogalacturonan II (RGII). Some pectins also contain non-carbohydrate domains, such as protein or ferulic acid. This presentation summarize two examples of pectin as an ingredient to promote health using encapsulation technology by pectin hydrogels and as an effective emulsifyier. The HG region of pectin was charge modified by pectinmethylesterase to create low or high methoxyl pectin and used to encapsulate hydrophilic and hydrophobic addenda as well as live probiotic cells via calcium chloride ionization. The encapsulation efficiency was high and release rate under gastrointestinal conditions was low for indomethacin or curcumin. The stability and in vitro release of Lactobacillus. FD L. casei FD L. casei encapsulated in charge modified pectin was evaluated. Beads maintained integrity under simulated gastrointestinal fluid, and no release was observed. The wet beads maintained viability of the encapsulated L. FD L. casei for 2 weeks, but nearly all flora died after 4 weeks storage. However, the dried beads of L. FD L. casei were viable for the 42 day study. When fed to rats on low fat or high fat diets, non-encapsulated and pectin encapsulated probiotic supplementation led to compositional changes in the gut microbiota in HF-fed rats. Gut barrier functions improved, local and systemic inflammation improved by non-encapsulated probiotics. Glucose tolerance improved by encapsulated probiotic treatment. Charge modified pectin hydrogels are potentially good vehicle for colon-targeted delivery carrier for probiotics. Emulsifying activity of pectin is attributed to protein content and quality, ferulic acid, hydrophobic domains, and charged domains. Sugar beet pectin was fractionated by sequential alcohol precipitation and emulsion were prepared with rice bran oil. Fractions precipitated at lower alcohol levels had improved emulsifying activity, higher surface hydrophobicity, and smaller particle size near 2 ±0.3 µm and low ζ-potential of -34±3 mV. Fractions varied in protein content, surface hydrophobity and glycome profile. Glycan-directed monoclonal antibodies response showed intense binding for homogalacturonan and rhamnogalacturonan I/ arabinogalactan epitopes.
