The goal of this project is to develop a quantitative non-isotopic assay for the detection of both single- and double-stranded forms of DNA in an agarose gel-based assay. This novel alternative would be an invaluable, safe option for research settings involving undergraduate students, researchers with limited experience, or without access to radioactive materials. While many methods exist for the detection and quantification of either single- or double-stranded DNA even in an gel-based assay, this alternative would relate both single- and double-stranded DNA concentrations to SYBR Gold signal intensity. A purification protocol was developed to isolate a pure, stable linearized DNA substrate free of undesirable DNA or extraneous salt contaminants from plasmid DNA preparations. The observed single- and double-stranded DNA signal intensity after SYBR Gold staining appears to be unique yet linear with respect to each DNA species. Further research is being conducted to determine the limit of detection, limit of quantification, and range of linearity for this method.
