As a food component, soluble non-digestible plant polysaccharides are generally recognized to function as dietary fiber and as prebiotics, and there is increasing recognition for immunomodulatory function. Pectin’s intrinsic chemistry/structure defines its utility as a food hydrocolloid and nutraceutical (e.g., “PectaSol-C”). We are analogously investigating arabinoxylan compositions prepared from rice bran fiber, particularly to determine structural features that may contribute to modulating innate immune responses at the intestinal epithelial barrier. We hypothesize that increased consumption of rice bran fiber with improved bioavailability of arabinoxylan (i.e., as a functional food or nutraceutical) can beneficially modulate inflammation in the intestinal mucosa. This presentation will highlight results to 1) characterize rice bran arabinoxylan compositions prepared by acid and enzyme hydrolyses in terms of their size/compositional profiles, 2) determine their activity to modulate TJ protein levels in T84 colonocytes, and 3) assess their ability to modulate response by T84 cells grown as confluent monolayers to pro-inflammatory cytokines.
