Nitrosamines are a series of organic nitroso compounds having the general form of R2NNO (R = alkyl and aryl). They have been found in the human environment such as foods, tobacco and rubber products. Recently, nitrosodimethylamine (NDMA) was detected as an impurity in several commonly used drugs for blood pressure and heartburn in US. It has raised great concerns due to the well-known carcinogenic effect of nitrosamines. The activation of nitrosamines by the heme-protein cytochrome P450 is required to produce highly reactive organic species, exerting their carcinogenic effects. The interactions of nitrosamines with heme proteins can be classified into Type I (nitrosamines binding to protein pocket sites) and Type II (nitrosamines binding direct to heme metal centers). To study the Type II direct interaction, we have prepared and characterized two six-coordinate and one five-coordinate nitrosodiethylamine (L) manganese porphyrin complexes of the forms, [(por)Fe(L)2]ClO4 (por = TPP (tetraphenylporphyrinato dianion), T(p-Cl)PP (tetrakis(p-chloro)porphyrinato dianion) and [(OEP)Fe(L)]ClO4 (OEP = octaethylporphyrinato dianion), respectively. The molecular structures have been determined by single-crystal X-ray crystallography.
