Paclitaxel is a chemotherapy drug used to treat a variety of cancers including lung, ovarian, and breast cancer. The drug possesses many drawbacks including poor aqueous solubility and is thereby formulated in a surfactant containing Cremophor/ EL. Concentrations of the drugs accumulated in tumors are low and require increasing the dosage, and drug accumulation in healthy tissues can cause dose-dependent toxicity. A recently discovered sugar polymer (sugar poly(orthoester) for use in engineering of nanomaterial may offer controlled release of the drug in the tumor site by taking advantage of the Warburg effect, which demonstrated the inefficient metabolism of cancer cells results in an acidic microenvironment, a stimulus that triggers a pH-responsive polymer to release loaded content. The systemic administration of paclitaxel-encapsulated sugar poly(orthoester) nanoparticles increased biodistribution to tumors and improved treating efficacy.
