Biocompatible and biodegradable polymeric nanoparticles are ideal candidates for drug delivery. Traditionally, drug molecules are either physically encapsulated into the nanoparticle, which may cause unintentional early release, or the drugs are covalently linked to the polymer backbone, which may result in inefficient release in vitro/in vivo and different drug actions compared to free drugs. In my presentation, I will discuss two drug delivery platforms that our group is investigating to address these challenges. The first platform consists of a drug-loaded nanoparticle prepared through non-covalent crosslinking of a triblock polynorbornene bearing β-cyclodextrin (β-CD) side chains and targeting groups. This delivery system is designed to carry combinations of free drugs in specific drug-binding sites, while permeating cellular membranes. The second platform is composed of a novel water-soluble star polymer that is synthesized via a core-first approach by grafting from a γ-CD multifunctional initiator core. This star polymer functions as a water-soluble host for hydrophobic drugs, and can also form well-ordered, highly crystalline solid-state materials. The syntheses and characterization of these two platforms, and the corresponding drug release kinetics and cytotoxicity will be described.
