The discovery and development of novel reactions that focus upon the transformation of chemical functionality is a cornerstone of organic synthesis. Our research group has recently developed a new method to directly install sulfonamide units into aldehydic carbonyls under mild, non-acidic conditions. This development has allowed for the production of new compounds formed from heteroaryl aldehydes that have the potential for bioactivity. In our previously reported research, indole-3-carboxaldehyde was used as a representative heteroaryl aldehyde in order to investigate the role of the sulfonamide unit in creating a cytotoxic compound. It was discovered that a 4-chlorobenzenesulfonyl unit created the most cytotoxic product from an initial set of sulfonamides that was tested. In this investigation, we aimed to test whether varying the substituents located about the periphery of the indole core would affect the potency of the resulting compounds. Our progress towards the expansion of a library of bioactive <i>N</i>-benzyl sulfonamides formed with indole cores will be presented along with results from a cell viability assay to determine the potency of the new compounds.
