Our research group is devoted on exploring facile fabrication on dual-emission probe consisting of monolayer MoS2 quantum dots (M-MoS2 QDs) and L-cysteine-conjugated boron-dipyrromethene (L-Cys-BODIPY) molecules for ratiometric sensing of biothiols and thiol product-related enzyme reactions. Self-assembled of L-Cys-BODIPY on the surface sulfur vacancys sites resulted in the fluorescence quenching of the M-MoS2 QDs via acceptor-excited photoinduced electron transfer (PET). Meanwhile, the conjugated L-Cys-BODIPY molecules were self-quenched through π-π stacking between each BODIPY backbones. The presence of biothiols induced the liberation of L-Cys-BODIPY from L-Cys-BODIPY-conjugated M-MoS2 QDs (BODIPY-M-MoS2 QDs), leading to the fluorescence restoration of L-Cys-BODIPY(Scheme 1). Moreover, the quenching of the M-MoS2 QDs by biothiols was found to be more efficient than that by L-Cys-BODPIY owing to small steric hindrance. Thus, the fluorescence of the BODIPY-M-MoS2 QDs progressively redcued with an increase in the concentration of biothiols. On the basis of (i) thiol-induced PET quenching of the M-MoS2 QDs and (ii) self-quenching of L-Cys-BODIPY by intramolecular π−π stacking of BODIPY backbones, the BODIPY-M-MoS2 QDs were well-suited for ratiometrically sensing biothiols and imaging glutathione-related reactions in live cells. Besides, the present probe in combination with S-adenosylhomocysteine hydrolase (SAHH)-mediated hydrolysis of S-adenosylhomocysteine was capable of detecting as low as 1 U L-1 SAHH with a linear range of 1 to 900 U L-1. The integration of the present probe with glutathione reductase-mediated reduction of disulfide GSH provided high selectivity and sensitivity toward glutathione reductase with a liner quantification range of 1-850 U L-1. Finally, the present probe was used to monitor the level of parathion and malathion in enviromental water.
