Nanotechnologies such as metal oxide nanoparticles have been shown in previous research to be a promising form of chemotherapy against cancerous cell lines. CuO nanoparticles (CuONPs) in particular are interesting in their ability to cause production of reactive oxygen species (ROS) within cells and to induce DNA damage. Elevating the already highly oxidative environment can help lead to and/or initiate apoptosis. Cytotoxic effects induced by exposure to CuONPs include a rise in cellular Cu<sup>2+</sup> concentrations, leading to an increase in ROS production as well as DNA damage caused by oxidative stress. We looked to uncover more on CuO’s applications in animal systems as a viable cancer treatment by analysis of size-differential cytotoxicity as well as their role in genotoxic outcomes. CuONPs of two varying sizes were synthesized/obtained and characterized via AFM, FTIR, and UV/Vis spectroscopy. The IC50 values of both size particles were assessed with MCF-7 cells. We analyzed intracellular Cu<sup>2+</sup> concentrations by ICP-OES as well as oxidative stress levels by ROS production. DNA damage was assessed following 24 hrs incubation with CuONPs in the presence and absence of thymoquinone to control for ROS production; the results of these studies will be detailed.
