Selective delivery of therapeutic agents to tumors is a major goal in medicine to avoid side effects. Gold nanoparticles have emerged as promising drug-delivery vehicles that can be synthesized with designer characteristics on the bench top for targeting cancer cells. However, under physiological conditions, various proteins non-selectively bind to the nanoconstructs in the form of a “corona” that may obscure the targeting abilities. This talk will describe a live-cell imaging assay that can differentiate between targeting and non-targeting nanoconstructs based on their single-particle dynamics. We will describe how gold nanostar nanoconstructs with two types of ligands—similar in size and charge, but different in their binding affinities to biomarkers on cancer cells—exhibit distinct translational and rotational motion, even though both of these nanoconstructs are surrounded by similar protein coronas. We will discuss how dynamics information can be used to inform how different nanoparticle characteristics—such as particle size, shape, and surface chemistry—may affect their potential as delivery agents by evaluating their efficacy under realistic conditions.
