Lumazine is a known photodynamic cancer therapy agent with an unknown mechanism of operation. The mechanism can occur via an excited-state electron transfer (type 1 cell death) and/or a formation of singlet oxygen (1O2) (type 2 cell death). In this research, we present a method for the determination of the 1O2 quantum yield in aqueous solutions using 2,5-diphenyl-3,4-isobenzofuran or 2,2,6,6-tetramethyl-4-piperidone as singlet oxygen traps and lumazine as the sensitizer. With the determined methodology the dependence of the 1O2 quantum yield of lumazine on oxygen concentration and lumazine concentration is measured. This work is completed in order to identify the contribution of 1O2 on the mechanism of cell death from irradiated lumazine.
