The Pesticide Analytical Manual is used for pesticide residue testing in non-fatty foods according to the Luke extraction. This extraction procedure utilizes liquid-liquid partitioning by the addition of petroleum ether and dichloromethane to the acetone sample filtrate. The principle of this procedure is the transfer of pesticide residues from the acetone extract (the aqueous layer) to the organic layer of dichloromethane and petroleum ether. The dichloromethane is finally reconstituted by addition of petroleum and acetone to the sample using a steam bath to obtain the sample extract in acetone. This extract is then cleaned using a tandem solid phase extraction (SPE) and graphitized carbon clean up procedure prior to analysis by mass spectrometry. This procedure is carried to determine if raw agricultural commodities comply with the Environmental Protection Agency’s standards, published in the Code of Federal Regulations (CFR) title 180, to ensure public health safety.
