While current treatments for burn-related infections occlude wounds, gauze and similar materials are not biodegradable and thus necessitate repeated replacement. In solution blow spinning (SBS), biodegradable polymer solutions are sprayed with an airbrush onto a wound. Here, we adapt SBS to create antimicrobial dressings by combining a blend of poly(lactic-co-glycolic acid) and poly(ethylene glycol) (PLGA/PEG) with silver ions (Ag+), an antimicrobial agent. PLGA/PEG/Ag dressings present numerous advantages over conventional approaches: (1) Minimal wound contact for decreased likelihood of pathogen transport. (2) Intrinsic adhesive properties for elimination of an additional layer of skin-irritating adhesive or gauze. (3) An antimicrobial agent in Ag+ that carries a relatively low minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The release profile of a specific PLGA/PEG/Ag composition was shown to initially release Ag+ in a burst fashion followed by 100% release after approximately 14 days, and at a concentration sufficiently antimicrobial but not strongly cytotoxic to cells in the wound. Addition of Ag+ resulted in lower Young's Modulus and ultimate tensile strength, as well as an unaffected wound closure adhesion strength. Durable polymer-scab hybrid scaffolds in porcine partial thickness wound healing studies indicated improved adhesiveness in PLGA/PEG/Ag materials as compared to Tegaderm, a polyurethane-based film often used in clinical settings. The combination of a favorable Ag+ release profile with a unique blend of biodegradable polymers that provide for a body temp temperature meditated adhesive event, establishes blowspun PLGA/PEG/Ag fibers as a potential burn wound dressing alternative.
