The number of people affected annually by bacterial pathogens represents a growing problem. Modern medicine utilizes antibacterial drugs to combat bacterial pathogens, but bacteria have the ability to evolve and render the medicine useless. Therefore, research is necessary to discover new antibacterial drugs to account for antibiotic resistance. Some species of bacteria (e.g. <i>Pseudomonas aeruginosa) </i>release chemicals called autoinducers (e.g. N-acyl homoserine lactones) that are detected by other bacterial cells in the population; this process is called quorum sensing. Bacteria use quorum sensing to detect population density and control gene expression. Once the concentration of autoinducers passes a certain threshold, they activate new genetic expressions such as bioluminescence or the formation of a biofilm. A biofilm is a polysaccharide coating created by bacteria to aid in protection and communication required for virulence. For example, pneumonia is a disease heavily reliant on the formation of a biofilm. Certain marine organisms have developed natural products that naturally disrupt the quorum sensing ability of bacteria by nullifying the autoinducers. One such example are the fimbrolides, butenolide natural products isolated from Australian red algae, which function as N-acyl homoserine lactone antagonists. Previous research has shown that shifts from the butenolide heterocyclic core to other heterocycles is capable of increasing the ability to disrupt quorum sensing. However, additional heterocylces remain to be explored. Progress towards the synthesis and biochemical evaluation of novel thiazolidine-2-thione fimbrolide analogs will be detailed. Synthesis of the heterocycle via Pd-catalyzed cyclization and subsequent acylation will be detailed. Antibacterial properties of selected compounds will also be presented.
