In recent years, the development of new pharmaceuticals has garnered much interest and despite many major advances in the field, one of the main challenges remains drug delivery. Herein we describe the use of cyclophosphazenes as the core of a drug delivery system. A hydrophilic phosphazene-based system was synthesized by substituting three equivalents of tetraethyleneglycol monomethyl ether (TEGME) onto the phosphazene trimer ring ([PCl<sub>2</sub>N]<sub>3</sub>). This now hydrophilic core can be furthered substituted with highly active pharmaceuticals, targeting agents, as well as imaging moieties. We are currently investigating the efficacy of substituting fluorescent imaging agents, such as coumarins, onto the previously synthesized phosphazene system. Coumarins have been widely used in the fields of chemistry, biology, and medicine and contain unique photophysical properties including high quantum yield as well as high stability and biological compatibility. Preliminary studies have shown that a variety of coumarins have been successfully substituted onto the phosphazene trimer ring. Results from these studies have been characterized via fluorescent spectroscopy as well as multinuclear NMR studies.
