The Emerald Ash Borer (EAB) has caused considerable damage to the North American ash (<i>Fraxinus spp</i>.) tree population and recently started attacking ornamental white fringe trees (<i>Chionanthus virginicus</i>). Previous research has shown that Manchurian Ash trees which is native to Asia, elicit a strong defense response against EAB and defend themselves from EAB due to its co-evolutionary history. The trees produce a coordinated response against the insect largely aligning with a systems biology model. Comparative metabolite profiling of trees, although quite useful requires complex analytical and statistical work-flow partly to address between sample variability. To date, several studies catalogued metabolites from ash trees, however, no information is available on white fringe tree metabolites. In order to understand the differences in constitutive and inducible metabolites between ash and white fringe tree, thirty-two trees were used in this study. The trees were either droughted and inoculated with (EAB) eggs or used as a control tree. We performed methanolic extraction of phloem tissue and catalogued metabolites using liquid chromatography – tandem mass spectrometry (LC-MSn). Metabolites were identified using authentic standards, fragmentation and information from literature. A detailed strategy and findings will be outlined in the presentation.
