Recent years have seen an explosion of interest in targeting RNA with small molecules from both academia and industry.  However, methods to identify, characterize, and study small molecules that bind to RNA are in their infancy compared to protein-binding inhibitors.  This presentation will detail my group's efforts to identify, characterize, and develop small molecules that bind to structurally and functionally diverse RNA targets.  By developing a small molecule microarray (SMM) screening platform, we can rapidly screen and profile a collection of ~40,000 druglike small molecules against an RNA of interest.  The presentation will describe efforts toward characterizing small molecules that are identified as hits from these screens, including inhibitors of oncogene expression and transcription. An an emphasis is placed on structural characterization of nucleic acid-ligand complexes, including our our group's efforts to develop structure-guided design techniques for RNA targets where we have obtained atomic resolution structures of ligands in complex with the target.
