α4/7-Conotoxin LvIA is from a family of selective inhibitors of acetylcholine receptors, some of which are linked to neurological diseases such as dementia, schizophrenia, and epilepsy. In order to study the function and application of this class of peptides as potential drug therapies, we are working towards an on-resin total synthesis of the native and misfolded structure of Conotoxin LvlA. Conotoxin has been sequenced by on-resin methods; however, the formation of the disulfide linkages are low in yield. These disulfide linkages play an important role in the folded structure and stability of proteins, so our is group interested in developing different cysteine protecting groups for selective disulfide formation in peptides in order to expand the peptide methodology tool box.
