Cells in the body constantly produce and degrade radicals called reactive oxygen species (ROS). Cancer cells have higher ROS levels than non-cancerous cells, and adaptation to these oxidants has been hypothesized to contribute to chemoresistance and cancer cell proliferation. Cancer cells have also been shown to exhibit altered patterns of endocytosis, but the possible link between oxidative stress and endocytosis has not been thoroughly investigated. Here we examine whether adaptation to oxidative stress alters endocytic behavior in MCF-7 breast cancer cells. We induce oxidative stress in cells using hydrogen peroxide in 24 hour and 7 day treatments, and compare resulting changes in the uptake of the fluorescent dye rhodamine 123 and of PLGA (poly lactic-co-glycolic) nanoparticles. This study will help to determine whether the effects of oxidative stress on endocytosis can be generalized across cell lines. It is also clinically relevant as levels of ROS could warrant inclusion in pharmacokinetic models.<br/>
