Sulfonamide functional groups are important within the area of drug discovery and development as isosteres of carboxylic acids, as noted by the number of pharmaceutical and bioactive compounds containing sulfonamide units. As a general class of therapeutic agents, N-alkyl and/or N-aryl sulfonamides have displayed a broad range of profiles that include antibacterial, anti-inflammatory, and anticancer activities. The incorporation of sulfonamide units to molecular scaffolds can lead to the development of a library of analogs to test for bioactivity, and thus, is a worthy goal within the realm of molecular synthesis. During our investigation of the incorporation of sulfonamide units to hindered alkyl halides under mild conditions, we have recently developed a new reaction that utilizes 9-bromofluorene as a hindered alkyl halide substrate in order to produce a library of N-sulfonyl analogs to test for cell viability. Our progress towards the development of a library of analogs by varying the electronic and steric properties of the N-sulfonyl unit on the fluorene scaffold, along with cell viability studies, will be presented.
