Intratumoral treatments offer an attractive approach to modulation of tumor microenvironments and activation of intratumoral immune mechanisms.  However, many current approaches are limited by poor intratumoral residence time of the candidate drugs.  This presentation will summarize our results using TransCon (short for transient conjugation) technology to achieve sustained, long-lasting intratumoral release of a TLR 7/8 agonist.  A single dose of TransCon TLR7/8 Agonist mediated a sustained local release of unmodified resiquimod, resulting in potent anti-tumor effects.  Importantly, TransCon TLR7/8 Agonist associated with significantly lower systemic proinflammatory cytokine induction when compared to an equivalent dose of unconjugated resiquimod, suggesting improved tolerability of the treatment.  TransCon TLR7/8 Agonist represents a new approach to intratumoral therapies and may overcome the challenges of existing approaches by providing potent anti-tumor responses while reducing adverse events related to systemic drug exposure.
