Benzoboroxoles are cyclic boronic acids that have shown impressive biological activities and this category of compounds have gained prominence with the approval of two drugs tavaborore (for the treatment of onychomycosis) and crisaborole (for the treatment of atopic dermatitis). Our group has been involved in the preparation of novel functionalized benzoboroxoles via simple methodologies amenable for scale up. We have utilized reactions such as aldol reaction, Baylis-Hillman reaction, Passerini reaction, allylation reaction, click reaction, among others for the efficient synthesis of diverse array of benzoboroxoles. We have also been able to accomplish further functionalization of the benzoboroxoles in an effort towards establishing the reactivity profile of these molecules. This presentation will focus on our recent efforts in this topic.
