In this study, we developed a series of thioether-based amphiphilic ionic liquids via the thiol-yne click reaction. We meticulously study their structure-property-activity relationships, developed via systematic variation of the distinct structural domains (viz. cationic headgroup, linker, and hydrophobic chains) for the optimized delivery of DNA into eukaryotic cells. The correlation between these amphiphilic ionic liquids’ structures and the magnitude of their transfection activity in vitro and toxicity will be reported. Furthermore, these cationic lipids collapsed into macroscopically hydrophobic phases by replacing their standard chloride anions with (CF3SO2)2N-. Indeed, clear and explainable relationships between the melting points of the products and the nature of each cation and anion are manifestly apparent, providing key insights into structural features that are consistent with ionic liquids that are simultaneously hydrophobic and low-melting.
