Copper ions have been implicated to play an important role in the angiogenesis process, formation of new blood vessels, which is high in metastatic breast cancer. One strategy being developed towards the treatment and prevention of breast cancer metastasis is to introduce drugs which deplete copper ions, thereby preventing the spreading of the cancerous cells. However, it is important to know the copper concentration in order to not deplete all the copper needed for normal healthy living cells, so that the appropriate dosage can be used with as little side effects as possible. We have developed a low cost nanoparticle-based localized surface plasmon resonance (LSPR) point of care assay to detect copper ions in biological samples in vitro. After reducing the cellular Cu2+ ions to Cu+ ions using ascorbic acid, a ‘click’ reaction is carried out to covalently couple a dye to the surface of a gold nanoparticle array. The dye-conjugated nanoparticles show a large shift in LSPR frequency particularly when the molecular resonance of the dye is overlapped with the LSPR of the nanoparticles. These shifts in LSPR frequency are also shown to be dependent on copper ion concentration, since copper acts as a catalysis for the conjugation of the dye to the gold nanoparticles. The assay shows promise towards the simple, rapid, and quantitative detection of copper ions in biological samples.
