Reactive oxygen species (ROS) such as superoxide anion radical (O2-), hydrogen peroxide (H2O2), singlet oxygen (1O2), and hydroxyl radical (.OH) are known to exist in all aerobic cells at low concentrations and acts as an intracellular signaling molecule. However, local overproduction of ROS in cells have been recently implicated in pathological conditions such as cancer. This overproduction induces oxidative damage at cellular level and plays a pivotal role in progression of cancer. Inspired by the unique ROS overproduction in cancer cells, the use of polymeric micelle-based drug carriers that dissociate to release drugs in response to elevated ROS levels has attracted growing attention to achieve cancer-specific drug delivery. Here, we report a library of polymeric micelles bearing thioether groups with different oxidation sensitivities. The H2O2-sensitive dissociation of the micelles could be finetuned by changing the substituent groups attached to the sulfur atom of thioether groups. Furthermore, the toxicity of the doxorubicin (Dox)-loaded polymeric micelles were evaluated in human liver cancer HepG2 cells and human umbilical vein cells (HUVECs).
