The fungal proteome has evolved to allow fungi to respond to the challenges of nutrient limitation and environmental conditions. Among the major exocellular enzyme components of the fungal proteome are phosphohydrolases. In Penicillium spinulosum cultures, the major phosphohydrolase activities detected in media filtrates are the phosphohydrolases, acid phosphatase (AP), bisphosphodiesterase (BP), and phosphorylcholine phosphodiesterase (PCPD) with AP activity surpassing BP and PCPD activities by several orders of magnitude in a modified Raulin Thom (mRT) medium. Our past investigations showed that P. spinulosum cultures grown in mRT media containing one-tenth of the normal phosphate with a ten-fold increase in iron(II) supplementation, had high acid phosphatase activity in 3-7 day cultures. This contrasts with the fungal cultures grown in control modified Raulin Thom media, where the AP activities peaked at 13 days. We have isolated exocellular AP proteins from both media. After chromatography and electrophoresis, selected AP protein bands from SDS PAGE were submitted for protein sequencing. The results of proteomic analyses of acid phosphatases from both regular and phosphate-limited/iron-supplemented mRT media and comparisons of their structures will be presented.
