In this poster, we present new polymerase chain reaction (PCR) high resolution melt (HRM) assays to detect and identify the pathogens Escherichia coli, Bacillus cereus, Clostridioides difficile, and Vibrio parahaemolyticus by their unique melt temperatures. All are human gut species that can cause infection when contaminated food is consumed or antibiotics alter the gut microbiome. The Centers for Disease Control and Prevention (CDC) has reported outbreaks of the foodborne pathogens V. parahaemolyticus and E. coli over the last several years. V. parahaemolyticus has infected imported fresh crab meat and raw shellfish and E. coli has infected bison, ground beef, flour, and Romaine lettuce. Developmental validation results using ATCC DNA including reproducibility, repeatability, specificity, sensitivity, and robustness will be presented. Testing with cultured pathogens and contaminated food products will also be presented.
