This study evaluated the phytochemical screening, antimicrobial activities as well as isolated and characterized the bioactive compounds extracted from the seeds of Piper guineense. The seeds were extracted by cold maceration with n-hexane, ethyl acetate and water. The phytochemical analysis conducted on the extracts using standard methods revealed the presence of alkaloids(7.68%), flavonoids(26.00%), saponins(9.80%),cardiac glycosides(1.50%), tannins(0.01%), terpenoids (50mg/g), steroids (13.5mg/g) and oxalate 1.215mg/100g. Antimicrobial screening and the minimum inhibitory concentrations (MIC) were also conducted on the three extracts using Agar broth dilution methods. The test organisms included four bacteria; Bacillus sp., Escherichia coli, Staphylococcus aureus and Streptococcus faecalis as well as two fungi isolates; Candida albicans and Aspergillus flavus. The largest zone of inhibition (32.5mm) was obtained with n-hexane extract against Staphylococcus aureus. The seed extracts of Piper guineense demonstrated notable antimicrobial potentials though below the positive control. The ethyl acetate extract exhibited the greatest antimicrobial activities against all the tested organisms followed by the n-hexane extract and then the water extract. Two compounds (major) 1-[5-(3,4-Methylenedioxyphenyl)-2-pentenoyl] pyrrolidine and (minor) 4,5-dihydro-21-methoxy piperine. were successfully isolated (using column chromatography and thin layer chromatography) and characterized (using 1H-NMR, 13C-NMR, COSY, HSQC and HMBC) from the seed extracts of Piper guineense.
