Cu (I) catalyzed Huisgen 1, 3-dipolar cycloaddition has been elegantly used in DNA modification. Our lab has been interested in the synthesis of boronic acid-modified DNA. In such a case, Cu(I)-mediated reactions pose a problem of stability to the boronic acid moiety. Herein we report for the first time the chemical synthesis of boronic acid-modified DNA using a copper-free click reagent.  This method can be used for the chemical synthesis of boronic acid-modified DNA aptamers for sensing and detection applications.
