Lab-scale purifications of diagnostic and therapeutic oligoribonucleotides and oligodeoxynucleotides often employ monodisperse porous resins. Diffusive mass transfer in these resins restricts resolution which and thus limits purification yields. These resins also exhibit limited flow ranges, due to restricted operational backpressures that lowers theoretical throughput.Use of polymeric monoliths coated with functional NanoBeads (latexes), like non-porous resins, produces narrow peak widths, supports chromatography over wide temperature and pH ranges, and support higher throughput as well as increased tolerance to high sample loads while still offering pH selectivity control. Here we present how combining NanoBead (Latex) technology with polymer monoliths produces oligonucleotide purification phases supporting improvements to both resolution and loading capacity when compared to conventional and monodisperse porous resins.
