Phosphatidylethanol (PEth) is a long-lived lipid metabolite of ethanol. Its levels in the circulation correlate with the amount of ingested alcohol up to two weeks after consumption. PEth is currently extracted from red blood cells and is measured by HPLC. To develop a better method of detecting PEth, we have prepared an amino modified PEth derivative and related phosphatidylethanolamine derivatives. Amino-PEth was used as an intermediate in preparation of fluorescent and biotinylated probes, as well as a substrate for the preparation of lipid-modified proteins. The protein�lipid conjugates were used as bait to screen a phage display library for sensitive and specific anti-PEth antibodies. We anticipate using anti-PEth antibodies to develop a simple colorimetric test (ELISA) for detection and quantitation of PEth in blood samples.
