Autooxidation kinetics of flavonoid compounds and flavonoid-containing berries were first tested in the presence or absence of reactive oxygen species. Next we observed the stronger inhibition of a-synuclein fibrillation by oxidation products of flavonoids than that by reduced forms. Flavonoid-containing berries afford more stable inhibition than the compounds. HPLC measurement verified the formation of three adducts of a-synuclein with flavonoids. It is surprised that only a weak covalent association of the flavonoid quinone with a-synuclein was observed by UV-Vis experiments. Formation of multiple hydrogen bonds are proved to be an important factor in promoting inhibitions on a-synuclein fibrillation. Flavonoids with three vicinal hydroxyl groups exhibited enhanced inhibition. We hypothesize that the hydrogen bonding increases hydrophilicity, produces hydrated a-synuclein molecules, restricts the conformational changes in the unfolded a-synuclein, and stabilizes the soluble flavonoid-modified a-synuclein monomers and oligomers.We acknowledge the support of NCSAMP and RISE programs of FSU.
