The immune system is charged with the recognition and elimination of foreign pathogens. The initiation of an immune response or the prevention of autoimmunity depends upon the ability of the B cell antigen receptor (BCR) to transmit signals that positively or negatively regulate B lymphocyte activation. Multivalent antigens generated through the ring-opening metathesis polymerization (ROMP), displaying ligands to the BCR and the coreceptor CD22, evoke a profound inhibitory effect on B cell activation. These synthetic probes alter signaling events and cellular processes regulated during antigen recognition.
