Multiplexed immunoassays are important for simultaneous detection of multiple biomarkers bringing reduced time, cost, and sample volume. Dengue fever presents in multiple serotypes and may be detected via the IgG and IgM from the host immune response. We present multiplexed detection of IgG and IgM using epi-fluorescence and  confocal laser scanning microscopy (CLSM). Conventional fluorescent dyes and quantum dots were compared in assays for anti-dengue IgM and IgG in the same well of an assay plate. The CLSM method allows the surface confined fluorescence to be detected. The epi-fluorescence microscope allows simultaneous excitation of quantum dots followed by the detection of light emitted over a wide spectral range using a spectral imaging add-on.  Multi well detection using enzyme linked immunosorbent methods gave good agreement with the new fluorescence methods.
