In addition to the canonicalribonucleosides, there are >100 enzymatically modifiedribonucleosides in tRNA and rRNA in all domains of life, with 15-25in any given organism.  Recent studies in yeast and human cells haverevealed a critical role for ribonucleoside secondary modificationsin the translational response of cells to toxic exposures.  Towardthe goal of understanding bacterial responses to the human innateimmune system in chronic inflammation, we have developed anLC-ESI-QTOF method to characterize and quantify the complete spectrumof ribonucleosides in microbial pathogens, beginning withHelicobacter pylori.  Initial studies to define the completespectrum of ribonucleosides have resulted in identification ofseveral previously unreported species.  After establishingdose-response relationships for exposure to microbicidal chemicalmediators of inflammation, exposure-induced changes in the spectrumof ribonucleoside secondary modifications were quantified byLC-ESI-QTOF.  The results provide insights into the role of RNAsecondary modifications in bacterial responses to inflammation.
