Repair of DNA interstrand cross-links is a process that is not well understood. It has been hypothesized that more than one repair pathway may be repsonsible to interstrand cross-link damage in mammalian systems. Since repair pathways can be linked to phases of the cell cycle, it was investigated here as to what cell cylcle abberations would occur in cells transfected with a site specific synthetically prepared interstrand cross-link in a plasmid substrate. The survival of cells was quantified by trypan blue exclusion and MTT mitochondrial function assays while cell cycle status was determined by Fluorescence Activated Cell Sorting.
