Iriomoteolide 1A is a 20-membered macrolide isolated from the dinoflagellate Amphidium strain HYA024.  It has been shown to exhibit potent cytotoxicity against human B lymphocyte DG-75 cells as well as the Epstein-Barr virus.  A highly convergent route to Iriomoteolide 1A is envisioned in which a non-selective aldol reaction unites aldehyde 1 and ketone 2 giving the entire carbon framework of Iriomoteolide 1A. Recent progress toward the total synthesis of Iriomoteolide 1A, including the synthesis of fragments 1 and 2 will be disclosed.
