The parnafungins (5) demonstrate broad spectrum antifungal activity with no antibacterial activity by inhibiting fungal polyadenosine polymerase.  We have developed a 5-step route to the hydroxymethyliodoxanthone (1) and a 3-step route to the pinacol arylboronate (2).  Suzuki coupling afforded 3. Reduction, mesylation and intramolecular SN2 reaction completed a short synthesis of the hexacyclic parnafungin A model 4.  Progress toward the synthesis of a fully functionalized left half and parnafungin A (5) will be described.
