An array based on thin film spots of DNA,metabolic enzymes and electroluminescent (ECL) ruthenium polymer ([Ru(bpy)2PVP10]2+)were used to screening metabolite-related toxicity of 11 toxic chemicals. Inthis approach, metabolic enzymes yielded products that can form covalentadducts with DNA. Then the array was transferred to a dark box apparatus forECL detection. A standard array with a representative range of metabolicenzymes was evaluated by estimating relative toxicity of these compounds interms of their ECL turnover rates R,the initial slope of ECL increase versus time per mass enzyme per mM substrate.Results from this array correlated well with those from the in vitro Comet DNAdamage assay and TD50 values for liver tumor formation in rodents. Resultsshow that the ECL array has the ability to provide valuable information for metabolites-relatedgenotoxicity, and is useful as a complementary chemical method to conventionalgenotoxicity bioassays.
