Cell surface carbohydrates act as receptors for a variety of protein ligands and thereby play a significant role in a wide range of biological processes. Therefore, carbohydrate binding interaction by protein has provided a starting point for the development of novel therapies, for protein and cell isolation and probing, and for targeted drug and gene delivery concept in biomedical research and applications. However, research using carbohydrates are restricted by synthesizing adequate molecules especially macromolecules since multivalency are normally involved in the carbohydrate recognitions. We report here oriented glyco-macroliand formation based on glycopolymer immobilization and glycosylated liposome immobilization strategies. First, an oriented glyco-macroligand formation and its glyco-affinity capturing of proteins will be demonstrated by combining O-caynate end-terminated glycopolymer with amine modified silica gel and glass slide, respectively. Second, a novel cyto-mimetic glyco-surface based on immobilized and glyco-modified liposome will be discussed. Briefly, PEG-triphenylphosphine-containing liposome was first chemoselectively immobilized onto an azide-modified glass slide surface and then further glyco-modified with azide derivated carbohydrate via Staudinger ligation. With this two-step Staudinger ligation strategy, we fabricated a 3 dimensional carbohydrate displaying that closely mimic cell surface carbohydrate environment. The intact liposomes immobilization and glyco-functionalization as well as their stability were confirmed with fluorescent releasing kinetics and lectin binding and AFM experiments.
