Cellulose derivatives such as hydroxypropyl methylcellulose (HPMC) arepromising raw materials for coatings or films for pharmaceutical applications. A case study is presented on hydrophilic biopolymerfor the preparation of oral controlled drug delivery systems (caps) [1]. Additivessuch as plasticizers, surfactants or lipids are frequently incorporated intobiopolymer matrices to produce high quality drug protective films. Formulationis a simple and elegant way to achieve specific properties. The studyinvestigates the influence of an hydrophilic plasticizer (polyol) and anhydrophobic excipient (fatty acid) on the surface properties of free HPMCfilms. On the basis of original atomic force microscopy (AFM) experiments,interplay between surface morphology, nanoadhesion and nanofriction aredetailed. The results clearly underline the strong dependence of coatingproperties on additive nature, concentration or water sensitivity and theinterplay with additive-biopolymer matrix compatibility.[1] A.
