We report polypeptide-based block copolymers combining a large range of experimental analysis in order to describe their self-assembly properties. One of our aim is to exploit the pH-sensitivity of polypeptides' secondary structure to manipulate the size and shape of the supramolecular structures formed by self-organization of these block copolymers In addition, the incorporation of magnetic nanoparticles in these assemblies bring the magnetization as another trigger that can be addressed in many different manners towards, attraction deformation, imaging or hyperthermia. The multi-responsive character of these polymersomes (T, pH, magnetic field) in the context of controlled drug delivery in vitro and in vivo will be addressed, demonstrating how a rational chemical design can impact a field as complex as cancer therapy and diagnosis.
