Peptide therapeutics by chemical synthesis represent an important class of Active Pharmaceutical Ingredients. Conventional organic solvent-based chemical syntheses, usually carried out in batch or semi-batch reactors, suffer from some issues: presence of poorly soluble by-products; deleterious side reactions; high organic solvent consumption; and large reaction time.The integration of membrane separation techniques with the reaction step addresses these problems. The application to peptide fragment condensation is presented.The key points of the proposed strategy are: incorporation of nanofiltration units with the reactor; continuous removal of undesired components from the mixture; recycle of solvent and reagents in excess; and continuous operating mode.The performances of the new strategy are presented for two industrial case studies, and compared with the conventional batch syntheses by means of techno-economical analysis. The Reactive Peptide Nanofiltration demonstrates advantageous in terms of costs and productivity.
