A simple HPLC method has been optimized for quantifying fatty acids with HPLC. Fatty acids extracted from soybeans were hydrolyzed and then tagged with the chromophore 2-bromo-2'-acetonaphthone for UV detection. The derivatization procedure was modified to omit an acetic acid co-solvent. Mobile phase composition and flow rate were optimized for peak resolution. Fatty acids were identified by comparison with commercially available standards derivitized with the acetonaphthone tag. Reproducibility was ensured by running sixteen replicate samples. Results showed similar fatty acid composition among the soybean samples.
