6�-(Hydroxymethyl)penicillin sulfones are potent and broad spectrum inhibitors of serine �-lactamases.  We have now prepared and evaluated a focused library of 2'�-substitute-6�-(hydroxymethyl)penicillin sulfones.  The 2'�-substituent can have substantial influence on the inhibitory activity and on the ability to penetrate Gram negative microorganisms.
