The architecture of synthetic macromolecular gene transfection agents is known to be significant in their efficacy, yet systematic architectural studies have been rare, in large part due to difficulties in the synthesis. Using a click cyclization technique developed in our labs, cyclic poly(ethylene imine) has been prepared from linear polyoxazoline precursors, along with exactly analogous linear poly(ethylene imine). The effect of architecture on the properties of these two polymers has been examined in detail.
