The death of dopaminergic neurons and accumulation of Lewy bodies (LB) are two hallmarks of Parkinson's disease (PD). In the brain of PD patients, elevated levels of Fe(III) were observed in region of LB where alpha-synuclein (a-syn) is a major component. Increased iron deposition in a-syn and neurodegeneration in the brain support the idea that possibly iron accumulation initiates dopaminergic degeneration in PD. The goal of this study is to probe kinetics and the mechanism of Fe(III)-catalyzed oxidation of dopamine, and the effect of a-syn on the reaction. In our previous work, we successfully separated short-lived dopamine oxidation species by using reverse-phase ion-pair chromatography and employing ion-trap mass spectrometry to detect the oxidation products in Fe(III)/ amyloid-beta mixture. The established LC-MS method was further applied to investigate the influence of a-syn on Fe(III)-catalyzed dopamine oxidation kinetics. Study here will shed light to the participation of iron in the pathology of PD.
